RESUMO

The aim of this work is to study the oregano essential oil (OEO) composition from Northwestern Argentinean regions and to evaluate its effect on the lactic starter cultures. The oregano used, Origanum vulgare var hirtum, was obtained from Andalgalá, Catamarca. The essential oil presented high amounts of α-terpinene (10%), γ-terpinene (15.1%), terpinen-4-ol (15.5%) and thymol (13.0%) as the main components. No negative effect on growth or metabolic activity of lactic acid bacteria Streptococcus thermophilus CRL 728 and CRL 813, Lactobacillus delbrueckii subsp. bulgaricus CRL 656 and CRL 468, and Lactococcus lactis subsp. lactis CRL 597 up to the maximum concentration (200µg/g) assayed was observed. No differences in the organoleptic characteristics of semi-hard cheeses flavored with oregano essential oil (200µg/g) and homemade cheeses flavored with oregano leaves were found. With respect to the microbiological quality of the products, neither enterobacteria nor mold and yeast were detected during ripening in essential-oil flavored cheese compared to control cheese (enterobacteria 2×103UFC/g) and cheese flavored with oregano leaves (mold/yeast 4×104CFU/g). Our results showed that the use of oregano essential oil and lactic starter culture considerably improved cheese quality.

El objetivo de este trabajo fue estudiar la composición del aceite esencial de orégano recolectado en el noroeste argentino y evaluar su efecto sobre algunos fermentos lácticos. El orégano recolectado correspondió a la especie Origanum vulgare var. hirtum proveniente de Andalgalá, Catamarca. En su aceite esencial (obtenido por arrastre con vapor de agua) se detectó principalmente α-terpineno (10%), γ-terpineno (15,1%), terpinen-4-ol (15,5%) y timol (13,0%). El aceite esencial no tuvo efecto inhibitorio (máxima concentración ensayada 200 μg/g) sobre el crecimiento ni sobre la actividad metabólica de Streptococcus thermophilus CRL 728 y CRL 813, de Lactobacillus delbrueckii subsp. bulgaricus CRL 656 y CRL 468, y de Lactococcus lactis subsp. lactis CRL 597. No se observaron diferencias en las características organolépticas de quesos semiduros aromatizados con el aceite esencial (200 μg/g) comparados con quesos artesanales aromatizados con hojas de orégano. Respecto de la calidad microbiológica de los productos, no se detectaron enterobacterias ni hongos o levaduras durante la maduración en los quesos aromatizados con el aceite esencial de orégano comparados con los quesos control, que presentaron desarrollo de enterobacterias (2 × 10³ UFC/g), y con los quesos elaborados con hojas de orégano, en los que hubo desarrollo de hongos/levaduras (4 × 10(4) CFU/g). Los resultados obtenidos demostraron que el uso del aceite esencial de orégano y del fermento láctico incrementó la calidad general de los quesos artesanales.

CONTEXT: The use of many traditional medicinal plants is often hampered by the absence of a proper biochemical characterization, which is essential to identify the bioactive compounds present in it. The essential oils (EOs) of three native species from the La Rioja province were analyzed: Lippia turbinata Griseb and Lippia integrifolia (Griseb.) Hieron (Verbenaceae), and Clinopodium gilliesii (Benth.) Kuntze (Lamiaceae). OBJECTIVE: The aim of this study was to evaluate their EOs antioxidant activity (AA) and their chemical composition. MATERIALS AND METHODS: EOs were analyzed by gas chromatography-mass spectrometry (GC-MS). To enhance the aqueous solubilization of the EOs, EO-water emulsions were prepared (concentration range of 0.1-6 mg mL(-1)). AA was determined using ABTS, DPPH, and peroxyl radical scavenging assays, as well as by the ß-carotene bleaching test. RESULTS: Piperitenone oxide was a major constituent in L. turbinata, pulegone and piperitenone oxide in C. gilliesii, and ß-caryophyllene in L. integrifolia. Lippia turbinata EO was the most active ABTS and DPPH radical scavenger (SC50 values of 0.40 ± 0.14 and 0.74 ± 0.08 mg mL(-1), respectively). Clinopodium gilliesii EO exhibited the highest hydrogen peroxide scavenging activity (SC25 value = 1.52 ± 0.27 mg mL(-1)). In the ß-carotene assay, L. turbinata EO was more effective at inhibiting lipid peroxidation than the other two oils (IC25 value = 0.15 ± 0.04 mg mL(-1)). CONCLUSION: Our results suggest that the AA observed can be justified by the presence of oxygenated monoterpenes, mainly piperitenone oxide. Finally, L. turbinata EO might be used as a safe natural antioxidant and food preservative in the food and cosmetic industries.

Gas chromatography-mass spectrometry (GC-MS) analysis revealed the major components in black cumin essential oils which were thymoquinone (37.6%) followed by p-cymene (31.2%), α-thujene (5.6%), thymohydroquinone (3.4%), and longifolene (2.0%), whereas the oleoresins extracted in different solvents contain linoleic acid as a major component. The antioxidant activity of essential oil and oleoresins was evaluated against linseed oil system at 200 ppm concentration by peroxide value, thiobarbituric acid value, ferric thiocyanate, ferrous ion chelating activity, and 1,1-diphenyl-2-picrylhydrazyl radical scavenging methods. The essential oil and ethyl acetate oleoresin were found to be better than synthetic antioxidants. The total phenol contents (gallic acid equivalents, mg GAE per g) in black cumin essential oil, ethyl acetate, ethanol, and n-hexane oleoresins were calculated as 11.47 ± 0.05, 10.88 ± 0.9, 9.68 ± 0.06, and 8.33 ± 0.01, respectively, by Folin-Ciocalteau method. The essential oil showed up to 90% zone inhibition against Fusarium moniliforme in inverted petri plate method. Using agar well diffusion method for evaluating antibacterial activity, the essential oil was found to be highly effective against Gram-positive bacteria.

The essential oil and methanol extract of northwestern Argentina medicinal plant Xenophyllum poposum, collected in Catamarca province, were investigated. GC and GC-MS analyses of the essential oil identified 56 compounds accounting for 92.9%. The main components of the oil were delta-cadinene (16.5%), 6-hydroxytremetone (14.7%), epi-alpha-cadinol (12.0%), alpha-cadinol (8.8%), gamma-cadinene (7.5%), 1-epi-cubenol (4.2%) and alpha-muurolene (3.0%). The essential oil exhibited antibacterial activities against five pathogenic strains as well as antifungal activities against two pathogenic fungi. The methanol extract showed antibacterial activity against two strains of Staphylococcus aureus and two pathogenic fungal strains. The main components isolated from the methanol extract were the antifungal 4-hydroxy-3-(isopenten-2-yl)-acetophenone, 6-hydroxytremetone, and tremetone. 6-Hydroxytremetone showed activity against all the fungal strains and one of the S. aureus strains assayed. Antioxidant and radical-scavenging properties of the methanol extract and essential oil were determined using the 2,2'-diphenyl-1-picrylhydrazyl assay and beta-carotene bleaching (BCB) test. The methanol extract and the essential oil showed, respectively, moderate and weak antioxidant activity when compared to butylated hydroxytoluene.

The essential oils of seven aromatic plants from central west Argentina were isolated by steam distillation and analyzed by a gas chromatography/mass spectrometry technique. The oils were screened for cytotoxicity and in vitro inhibitory activity against herpes simplex virus type 1 (HSV-1), dengue virus type 2 (DENV-2) and Junin virus (JUNV). The oils showed a variable virucidal action according to the virus. JUNV was the least susceptible virus in comparison with HSV-1 and DENV-2. The better relationship between cytotoxicity and inhibitory activity was observed for the essential oil of Lantana grisebachiii (Seckt.) var. grisebachii against DENV-2 and HSV-1 with IC50 (inhibitory concentration 50%) values of 21.1 and 26.1 ppm, respectively. This effect was specific since the selectivity indices (ratio cytotoxicity/virucidal activity) were > 23.7 and > 19.1 for DENV-2 and HSV-1, respectively. Furthermore, the oil from L. grisebachii was also an effective inhibitor of HSV-2 and acyclovir resistant variants of herpes virus. This study demonstrates the effective and selective inhibitory activity of the essential oil from Lantana grisebachii against HSV and DENV by direct virus inactivation.

The essential oils of seven aromatic plants from Córdoba, San Luis and San Juan Provinces (Argentina) were isolated by steam distillation and analysed by a gas chromatography/mass spectrometry technique. The oils were screened for cytotoxicity and in vitro inhibitory activity against herpes simplex virus type 1 (HSV-1), dengue virus type 2 (DENV-2) and Junin virus (JUNV) by a virucidal test. The oils showed a variable virucidal action according to the virus. The better relationship between cytotoxicity and antivirus action was observed with the essential oils of Heterothalamus alienus and Buddleja cordobensis against JUNV, with virucidal concentration 50% (VC50) values of 44.2 and 39.0 ppm and therapeutic indices (cytotoxicity to virucidal activity ratio) of 3.3 and 4.0, respectively. The inhibitory action was exerted by a direct interaction of virions with the oils. Virions inactivated with B. cordobensis and H. alienus essential oil were not affected in their ability to bind to the host cell. The therapeutic indices shown by these essential oils in toto were very modest, but given the complexity of their chemical composition the future identification of the precise active principle may allow the elimination of cytotoxic components and increase the selectivity of the effective compound.
